Modeling genetic lung disease using hPSCs

This project is in collaboration with Dr. Benjamin Raby, director of theĀ BWH Pulmonary Genetics Center, and Xiaobo Zhou.

Together, our labs have set out to functionally dissect regulatory variants at established lung disease-associated loci using RNA guided genome engineering and hESCs. We are generating isogenic hESC lines carrying lung disease associated mutations of interest using CRISPR/Cas9 genome editing.

We are building on work done in our lab on differentiating hESCs to NKX2.1+ lung progenitor cells. Our goal is to generate hESC-derived bronchial epithelium and demonstrate specific, distinct molecular deficits and corresponding cellular phenotypes in the differentiated cells.
